Enzymatic Synthesis of Amoxicillin with Immobilized Penicillin G Acylase
نویسنده
چکیده
The synthesis of amoxicillin with immobilized penicillin G acylase (PGA) in aqueous medium was investigated. The parameters studied were: time course of amoxicillin production, concentration of substrates: hydroxyphenylglycine methyl ester (HPGM) and 6-aminopeicillanic acid (6APA) and the e ect of enzyme (PGA) content and pH, under variable and constant conditions and temperature variations. In the study of two substrate concentration on amoxicillin production, impressive results were obtained for a 1/3 ratio of 6-amino penicillanic acid (6-APA) and hydroxyl-phenylglycine methyl ester (HPGM). The synthesis of amoxicillin was preferable at constant pH rather than a variable one. Other optimal conditions obtained were: enzyme concentration: 5 g/L with 100U, process time: 480 min and temperature: 35 C. The yield for amoxicillin synthesis under prescribed conditions showed up to 50%.

متن کاملSolid-liquid Equilibrium Data of Amoxicillin and Hydroxyphenylglycine in Aqueous Media
The enzymatic synthesis of amoxicillin is catalyzed by Penicillin G Acylase (PGA). As byproducts, hydroxyphenylglycine and alcohol are also formed from hydrolytic reactions and antibiotic synthesis, respectively. The design of this process should be directed to promote the synthesis reaction. At the same time, it is necessary to reduce the hydrolytic reaction of amoxicillin through its crystall...

An experimental design was applied to improve the reaction conditions for enzymatic synthesis of ampicillin from phenylglycine methyl ester (PGME) and 6-aminopenicillanic acid (6-APA), catalyzed by penicillin G acylase from E. coli immobilized on an agarose-glyoxyl derivative. The presence and magnitude of interactions between reaction variables were estimated using a 2 factorial design. A batc...

The catalytic properties of penicillin G acylase (PGA) from Escherichia coli in kinetically controlled synthesis of β-lactam antibiotics are negatively affected upon immobilization on hydrophobic acrylic carriers. Two strategies have been here pursued to improve the synthetic performance of PGA immobilized on epoxy-activated acrylic carriers. First, an aldehyde-based spacer was inserted on the ...
